Bioeconomic production of high-quality chitobiose from chitin food wastes using an in-house chitinase from Vibrio campbellii

Abstract Marine Vibrio species are natural degraders of chitin and usually secrete high levels chitinolytic enzymes to digest recalcitrant chitooligosaccharides. This study used an endochitinase ( Vh ChiA) from campbellii produce high-quality chitobiose crustacean chitins. The enzyme was shown be fully active stable over 24 h when BSA as additive. When different sources were tested, ChiA preferentially digested shrimp squid (?) chitins compared crab (?) did not utilize non-chitin substrates. overall yields obtained small-scale production using a single-step reaction 96% shrimp, 91% pen crab-shell Larger-scale yielded 200 mg chitobiose, with > 99% purity after desalting purification step preparative HPLC. In conclusion, we report the employment in-house produced chitinase effective biocatalyst rapidly convert food wastes in quantity quality suitable for use research commercial purposes. Chitobiose by this economical eco-friendly approach can easily scaled up obtain multi-gram quantities chemo-enzymic synthesis rare chitooligosaccharide derivatives long chain chitooligosaccharides, well preparation sugar-based functionalized nanomaterials.
